You do not have to uninstall the your current EarMaster 4.0 installation.

You should just install the updated version into the same directory as your current EarMaster 4.0 Installation.

latest version?

Go to the "Help" in EarMaster 4.0 and choose "About EarMaster 4".
Look for the text in gray: e.g. (build 391)
You do not have the latest version installed if your build code is lower than 489.
